Mitogen-activated protein kinase-activated protein kinase 5 (MAPKAPK5), also known as MK5 or p38-regulated/activated protein kinase (PRAK), is a novel member of p38 downstream Ser/Thr protein kinase family, with 471 amino acid residues, expressed ubiquitously in human tissues. The protein is characterized with conserved protein kinase domains I–XI.

生化/生理作用
Mitogen-activated protein kinase-activated protein kinase 5 (MAPKAPK5) plays a vital role in negative regulation of doxorubicin-induced apoptosis in human hepatocellular carcinoma cells (HCC). Reduced expression of MK5 is associated with colon cancerogenesis in humans. PRAK induces phosphorylation of heat shock protein 27 (HSP27) in vivo, in response to cellular stress. PRAK activity is regulated by the p38 pathway. PRAK plays an essential role in tumor suppression and Ras-induced senescence. Decreased expression of PRAK results in increase in mammalian target of rapamycin complex 1 (mTORC1) activity and thus contributes to tumor development. PRAK stimulates various cellular signaling process such as cell cycle, angiogenesis and neuronal plasticity. PRAK might acts as a therapeutic target for Alzheimer′s disease (AD) via interacting with receptor for advanced glycation end products (RAGE)-mediated cell signaling pathway induced by amyloid β (Aβ).
