PF-03491165 is a potent HMG-CoA reductase inhibitor (IC50 = 0.1 nM) with good hepatoselectivity (rat hepatocyte/myocyte cholesterol synthesis IC50 = 0.1 nM/444 nM). PF-03491165 exhibits superior plasma low density lipoprotein (LDL)-lowering efficacy than rosuvastatin and pravastatin in Guinea pigs in vivo (EC50 = 5.9, 13.5, 73.3 mg/kg, respectively via daily po) despite its low oral bioavailability in rats and dogs.
